Alignment

Alignment is the procedure that SBTengineŽ uses for allele assignment. SBTengineŽ aligns and compares your sequence files with the information available in the IMGT/HLA database. SBTengineŽ can do this because it "interprets" the electropherograms of your sequence files by basecalling. Some regions in the electropherogram of your sequence files may not be optimal which may lead to wrong basecalling. Wrong basecalling at non-polymorphic positions of the HLA gene results in a discrepancy between your sequence file data and the IMGT/HLA database. This is called an inconsistency. Any inconsistencies between your sequence files and the database are indicated and need to be addressed. SBTengineŽ leads you through all inconsistent positions and allows you to make the appropriate changes. All changes to the original sequence files of a sample are monitored and can be viewed with the "modified base" option.
