Editing
SBTengine analyzes the electropherograms of your sequence files and performs the basecalling. Basecalling is the interpretation of the peaks in the electropherogram. This determines which nucleotide belongs at which position. Some regions in the electropherogram of your sequence files may not be optimal which can lead to incorrect basecalls. SBTengine indicates and leads you through all incorrect basecalls. This allows you to interpret the electropherograms and edit the basecalling manually without the need to inspect all sequenced nucleotides. This saves a lot of work.
How to edit the base-calling?
1. Select the position you want to edit. You may either select a position by clicking on it with your mouse pointer or by following the list in the right pane of the sequence overview. The selected position is indicated with a blinking cursor in the electropherogram.
2. Enter the correct base-calling (A, T, C & G) using your keyboard and press Enter. NC-IUB codes for polymorphic positions are automatically set when entering multiple nucleotides at one position.
Example: when you sequentially enter a "G" and an "A" at a polymorphic postition, SBTengine will automatically display the NC-IUB code "R". You can also use the handy diagram shown below to determine the NC-IUB code. Go to the NC-IUB codes topic to learn more about this feature.
