In case there is no allele assignment, SBTengine® will show a list of crucial positions that you need to check. Check the list and edit the basecalling at the crucial positions according to the electropherogram. After each edited nucleotide the list will be updated. Crucial positions that are checked will change from bold to normal and will be placed at the bottom of the list. In most cases this action will result in an allele assignment.

 

In case there are no crucial position shown at all, there are possibly too many nucleotides that were not at all (as  N's) or ambiguously called. In a few cases, sequences may have insertions or deletions in the sequence that could not properly be called by SBTengine. A message will appear in the lower right corner to indicate the postion(s) of the insertion(s)/deletion(s). You may solve this by manually evaluating the sequences and edit accordingly as described in the Insertions/Deletions section.

 

On the other hand, you may have a new allele in you sample that is currently not yet known to the IMGT/HLA database. You can find more information on how to recognize and deal with new alleles in the 'New Alleles' section.
