Allele ratios

 

SBTengine reports the allele ratios for heterozygous samples in the top left corner of the electropherogram.

 

These ratios are based on the peaks at positions where the two alleles in the sample differ. At these positions the ratio of the peaks of one allele to the other is calculated. The mean of these ratios are shown in the upper left corner of the trace, and can be used to inspect the balance between the two alleles, or which allele has been sequenced by a particular GSSP. The ratios are calculated based on the alleles of the first genotype listed in the typing list.

 

For heterozygous sequences, the mean of the ratios for both alleles are shown. In the example, 51:49 is the signal ratio between the B*08:01 and B*39:01. For the B*08:01, the ratio 51 ranges from 40-64, and is based on peaks at 9 positions (n=9). Other ambiguities (in this case B*39:01:01:02L) are not displayed.

 

For homozygous sequences, e.g. GSSP, the fraction of the allele sequenced is shown. In a perfectly homozygous sample without any noise or traces of the other allele, a percentage of 100% is expected. In the example below the B*39:01 represents 93% of the signal. This value ranges from 83-98 over the 7 positions inspected (n=7).

 

Ratios are not shown when:

- No exact matching genotype is determined

- The region or sample is homozygous

- The sequence of one or two of the alleles is not known in the region covered by the trace
